Selank

Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro-Desmethyl)

Also known as: Selank, TP-7, Selanc

Overview

A synthetic analog of the naturally occurring immunomodulatory peptide tuftsin, developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. Selank is primarily recognized as an anxiolytic and nootropic peptide, but its relevance to sleep lies in its potent anti-anxiety effects and modulation of GABAergic neurotransmission. By reducing anxiety and normalizing stress responses without sedation, Selank helps resolve the anxiety-driven insomnia that affects a large proportion of sleep disorder patients.

Half-Life
~3-5 minutes (native); active metabolites persist for several hours
Typical Dose
250–750 mcg
Frequency
1-3 times daily; evening dose for sleep support
Cycle Length
2-4 weeks on, 2 weeks off

Administration

IntranasalSubcutaneous

Benefits

  • Reduces anxiety that causes insomnia and sleep-onset difficulty
  • Modulates GABA system without sedation, tolerance, or dependence
  • Improves sleep quality in stress-related and anxiety-driven insomnia
  • Enhances cognitive function and memory consolidation during sleep
  • Stabilizes mood and emotional regulation
  • Immunomodulatory effects supporting overall health

Side Effects

  • Nasal irritation (intranasal route)
  • Mild fatigue during initial adaptation
  • Headache (uncommon)
  • Generally very well tolerated with no reported dependence

Mechanism of Action

  • Allosteric modulation of GABA-A receptors to enhance inhibitory neurotransmission without benzodiazepine-like sedation
  • Inhibits enkephalin-degrading enzymes, prolonging endogenous anxiolytic opioid signaling
  • Modulates serotonin (5-HT) and dopamine metabolism for mood stabilization
  • Influences BDNF expression for neuroprotection and synaptic plasticity
  • Activates IL-6 and T-helper cell pathways for immune modulation (tuftsin analog activity)

Contraindications

  • Concurrent use of benzodiazepines or strong GABAergic drugs (potential interaction)
  • Active autoimmune conditions (immunomodulatory effects may be unpredictable)
  • Pregnancy or breastfeeding

Storage & Reconstitution

Store at 2–8°C (36–46°F) for up to 3 months, protected from light and heat. Intranasal solutions: refrigerate and use within 14-21 days.

Common peptide amounts:2 mg5 mg

Research Summary

Selank has been approved in Russia as an anxiolytic medication (nasal spray formulation) and has been studied in over 30 clinical and preclinical trials. Studies demonstrated anxiolytic effects comparable to benzodiazepines but without sedation, cognitive impairment, or dependence. Zozulia et al. showed that Selank normalized the sleep-wake cycle in patients with generalized anxiety disorder, with improvements in both sleep onset latency and sleep maintenance. Gene expression studies revealed modulation of 45 genes related to GABAergic and serotonergic neurotransmission. Its dual anxiolytic-nootropic profile makes it particularly valuable for individuals whose sleep disturbance stems from anxiety or cognitive hyperarousal.
